Leucyl transfer RNA synthetase changes during soybean cotyledon senescence.

چکیده
Previous work has shown the complement of leucyl transfer RNA (tRNA(Leu)) isoaccepting species present in soybean cotyledons to change during cotyledon senescence. Of the six tRNA(Leu) species resolved by Freon-column chromatography, the capacity of species 1-4 to accept leucine is decreased between 5 and 21 days of age. Parallel changes in tRNA(Leu) synthetase activity are reported here during the same time period. There is a reduction in the capacity of the synthetase from 21-day-old cotyledons to aminoacylate tRNA(Leu) 1-4, while the capacity to charge tRNA(Leu) 5-6 remains high. When tRNA(Leu) derived from 5-day-old cotyledons is maximally charged with the 21-day synthetase, the subsequent addition of synthetase from 5-day-old cotyledons promotes additional charging, primarily of species 1-4. These results suggest the presence of tRNA(Leu) "subspecies" within the individual peaks resolved on a Freon column, differing in synthetase specificity.

Isolation of an organ-specific leucyl-tRNA synthetase from soybean seedling.
The leucyl-tRNA synthetase activity from cotyledons of 4-day-old soybean seedlings has been fractionated into three components. One of these exclusively acylates two of the six tRNA(Leu) species present in this tissue. The remaining two enzyme fractions charge the other four tRNA(Leu) species equally well. Soybean hypocotyls appear to contain only the last two enzyme fractions.
متن کاملIsolation and partial characterization of temperature-sensitive Escherichia coli mutants with altered leucyl- and seryl-transfer ribonucleic acid synthetases.
Two temperature-sensitive mutants of Escherichia coli have been found in which the conditional growth is a result of a thermosensitive leucyl-transfer ribonucleic acid (tRNA) synthetase and seryl-tRNA synthetase, respectively. The corresponding genetic loci, leuS and serS, cotransduce with lip and serC, respectively. As a result of the mutationally altered leucyl-tRNA synthetase, some leucine-,...
متن کاملNucleic Acid and protein metabolism of senescing and regenerating soybean cotyledons.
An alternative to the leaf disk system for studies of the metabolism of senescence is described. The progress of senescence of soybean (Glycine max L.) cotyledons is arrested when the epicotyl is removed. Epicotyl removal at 16 or 17 days reversed the decline in nucleic acid, protein, and chlorophyll content in the cotyledon. Epicotyl removal at 18 days did not reverse the decline in the above ...
متن کاملA green-cotyledon/stay-green mutant exemplifies the ancient whole-genome duplications in soybean.
The recent whole-genome sequencing of soybean (Glycine max) revealed that soybean experienced whole-genome duplications 59 million and 13 million years ago, and it has an octoploid-like genome in spite of its diploid nature. We analyzed a natural green-cotyledon mutant line, Tenshin-daiseitou. The physiological analysis revealed that Tenshin-daiseitou shows a non-functional stay-green phenotype...
متن کاملPleiotropic phenotype of an Escherichia coli mutant lacking leucyl-, phenylalanyl-transfer ribonucleic acid-protein transferase.
A mutant of Escherichia coli that lacks leucyl-, phenylalanyl-transfer ribonucleic acid-protein transferase had diminished activities of L-phenylalanyl-transfer ribonucleic acid synthetase and tryptophanase, grew faster than its parent with aspartic acid as the sole nitrogen source, accumulated higher levels of enterochelin in the medium during iron limitation, and exhibited an abnormal morphol...
